with the MagCore® System
Total DNA purified by the MagCore Tissue and FFPE Tissue Kits is ideal for sensitive downstream applications such as PCR, Real-Time PCR, and multiple PCR. Users can count on its reliability to routinely operate total DNA extraction.
Two protocols have been devised and optimized for different sizes of tissues: 2 hrs for small samples/ 16 hrs for large samples.
Archived Formalin-Fixed, Paraffin-Embedded (FFPE) tissues are valuable samples to profile gene expression or study diseases. Amplifying DNA from scarce and low-quality FFPE tissue can be tricky.
The MagCore Genomic DNA FFPE process is designed for purification of total DNA from FFPE tissues with the MagCore instruments. It features One-Step Heating, to melt the paraffin and lyse tissue samples at the same time without applying harmful reagents such as xylene.
The surface area of the FFPE tissue sample measured as shown in examples. Sample amount of preparation can be 1-5 scrolls, each with a thickness up to 5μm. One FFPE scroll could be enough to analyze if the surface area is over 200 mm2.

Here we show the complete workflow of DNA extraction and purification from FFPE samples - starting with neutralizing paraffin and ending with the DNA purification.
The MagCore® Genomic DNA FFPE One-Step Kit is used with the MagCore Nucleic Acid Extractor.

MagCore-treated samples have mostly lower (better) CT values as compared to samples processed with a competitor's product, regardless if the 2-hour or 16-hour heating protocol is applied.
